🧬 Biomolecules, Polymers & Practical Chemistry

Biomolecules and polymer sciences bridge organic mechanisms with biochemistry and materials science. Advanced competitive examination topics include D/L sugar nomenclature, mutarotation kinetics & anomeric carbon configurations, amino acid isoelectric point (pI) electrophoretic migration, and step-growth vs chain-growth polymerizations.

1. 🍞 Carbohydrates: Monosaccharides & Disaccharides

1.1 Glucose (C6H12O6, an Aldohexose)

  • D/L Configuration: Determined by configuration of the lowest asymmetric carbon (C-5 in glucose). If −OH is on the right in Fischer projection implies D-Sugar.
  • Cyclic Structure & Anomers: Intramolecular nucleophilic addition of C-5 −OH to C-1 aldehyde gives 6-membered cyclic hemiacetal (α-D-Glucopyranose and eta-D-Glucopyranose).
    • α and eta anomers differ strictly in configuration at C-1 (Anomeric Carbon).
  • Mutarotation: Spontaneous change in specific optical rotation of an optically active anomer in aqueous solution until equilibrium is reached (+112∘→+52.7∘←+19∘).

1.2 Reducing vs. Non-Reducing Sugars

  • Reducing Sugars: Possess a free hemiacetal/hemiketal −OH on anomeric carbon $ o$ reduce Tollens' and Fehling's reagents (All monosaccharides like Glucose, Fructose; Disaccharides like Maltose, Lactose).
  • Non-Reducing Sugars: Anomeric carbons of both monosaccharide units are linked in glycosidic bond (e.g. Sucrose, where C-1 of α-D-Glucose links with C-2 of eta-D-Fructose).

2. 🧪 Amino Acids & Proteins

Amino acids possess both an acidic carboxyl group (−COOH) and a basic amino group (−NH2). In aqueous solution, they exist predominantly as dipolar Zwitterions (H3N+−CHR−COO−).

2.1 Isoelectric Point (pI) & Electrophoresis

The Isoelectric Point (pI) is the specific pH at which the amino acid carries zero net electrical charge and does not migrate in an electric field:

pI=pKa1+pKa22
  • At pH<pI: Solution is acidic $ o$ Amino acid exists as Cation (H3N+−CHR−COOH) implies migrates toward Cathode (−).
  • At pH>pI: Solution is basic $ o$ Amino acid exists as Anion (H2N−CHR−COO−) implies migrates toward Anode (+).
